不同发育时期香橼果实囊衣萜类物质代谢组学分析

        Metabonomic analysis of terpenoids for segment membrane of citron at different fruit development stages

          目的 以不同果实发育时期的香橼囊衣为材料,开展靶向萜类物质代谢组检测与分析,为解析不同果实发育时期萜类物质代谢途径提供参考。
          方法 对香橼4个果实发育时期(青果期、黄绿果期、绿黄果期、黄果期)的囊衣进行UPLC-MS/MS检测,并进行代谢物定性定量分析、样本质控分析、主成分分析、聚类分析、OPLS-DA分析及差异代谢物KEGG功能注释与富集分析。
          结果 从香橼果实囊衣中共检测到215种萜类代谢物,包括单萜90种、三萜51种、倍半萜49种、二萜16种、三萜皂苷4种、其他萜类5种,以柠檬苦素和诺米林的含量较高;在青果期的萜类物质积累相对较多,但在不同时期萜类物质的积累具有特异性;筛选出79个显著差异代谢物,不包括柠檬苦素和诺米林;单萜类和二萜类生物合成途径在4个发育时期的富集程度高,表现显著。
          结论 青果期香橼果实囊衣的萜类物质含量较高,单萜类和二萜类生物合成途径富集程度高。

           

          Abstract:
          Objective Targeted terpenoids metabolome detection and analysis were carried out to provide references for further analysis of terpenoids metabolism pathway in segment membrane at different fruit development stages.
          Method UPLC-MS/MS was used to detect terpenoids in segment membrane at four fruit development stages (Green, yellowish-green, greenish-yellow and yellow fruit stages), and the qualitative and quantitative analysis of metabolites, sample quality control analysis, principal component analysis (PCA), cluster analysis, OPLS-DA, KEGG functional annotation and enrichment analysis of differential metabolites were used to explore the differences of terpenoids metabolites at four fruit development stages.
          Result A total of 215 metabolites were detected, including 90 monoterpenes, 51 triterpenes, 49 sesquiterpenes, 16 diterpenes, 4 triterpenoid saponins, and 5 other terpenoids. The content of limonin and nomilin was relatively high. Terpenoids accumulated relatively more at the green fruit stage, but their accumulation exhibits specificity at different fruit development stages. A total of 79 significantly different metabolites were identified, not including limonin and nomilin. The enrichment degree of monoterpenes and diterpenes biosynthesis was high and significant at four development stages.
          Conclusion The content of terpenoids is relatively high at the green fruit stage, with a high degree of enrichment in the biosynthesis of monoterpenes and diterpenes.
